Phosphatidylinositol 4,5‐bisphosphate (PI4,5P2) is a key lipid signaling molecule that regulates a vast array of biological activities. PI4,5P2 can act directly as a messenger or can be utilized as a precursor to generate other messengers: inositol trisphosphate, diacylglycerol, or phosphatidylinositol 3,4,5‐trisphosphate. PI4,5P2 interacts with hundreds of different effector proteins. The enormous diversity of PI4,5P2 effector proteins and the spatio‐temporal control of PI4,5P2 generation allow PI4,5P2 signaling to control a broad spectrum of cellular functions. PI4,5P2 is synthesized by phosphatidylinositol phosphate kinases (...) (PIPKs). The array of PIPKs in cells enables their targeting to specific subcellular compartments through interactions with targeting factors that are often PI4,5P2 effectors. These interactions are a mechanism to define spatial and temporal PI4,5P2 synthesis and the specificity of PI4,5P2 signaling. In turn, the regulation of PI4,5P2 effectors at specific cellular compartments has implications for understanding how PI4,5P2 controls cellular processes and its role in diseases. (shrink)

Subjects were exposed to sentences containing “direct” and “indirect” uses of names and definite descriptions. On a subsequent recognition test incorrect rejections tended to be of sentences involving indirect uses, and false alarms to sentences involving direct uses. This finding is contrary to the predictions of models that suggest indiscriminate substitution of names for descriptions, as do those of Anderson and Bower, and Rumelhart and Norman. The meaning of most words in language depends on their context. Understanding how the human brain extracts contextualized meaning, and identifying where in the brain this takes place, remain important scientific challenges. But technological and computational advances in neuroscience and artificial intelligence now provide unprecedented opportunities to study the human brain in action as language is read and understood. Recent contextualized language models seem to be able to capture homonymic meaning variation (“bat”, in a baseball vs. a vampire context), as (...) well as more nuanced differences of meaning—for example, polysemous words such as “book”, which can be interpreted in distinct but related senses (“explain a book”, information, vs. “open a book”, object) whose differences are fine‐grained. We study these subtle differences in lexical meaning along the concrete/abstract dimension, as they are triggered by verb‐noun semantic composition. We analyze functional magnetic resonance imaging (fMRI) activations elicited by Italian verb phrases containing nouns whose interpretation is affected by the verb to different degrees. By using a contextualized language model and human concreteness ratings, we shed light on where in the brain such fine‐grained meaning variation takes place and how it is coded. Our results show that phrase concreteness judgments and the contextualized model can predict BOLD activation associated with semantic composition within the language network. Importantly, representations derived from a complex, nonlinear composition process consistently outperform simpler composition approaches. This is compatible with a holistic view of semantic composition in the brain, where semantic representations are modified by the process of composition itself. When looking at individual brain areas, we find that encoding performance is statistically significant, although with differing patterns of results, suggesting differential involvement, in the posterior superior temporal sulcus, inferior frontal gyrus and anterior temporal lobe, and in motor areas previously associated with processing of concreteness/abstractness. COVID-19 is a highly contagious infection with no proven treatment. Approximately 2.5% of patients need mechanical ventilation while their body fights the infection.1 Once COVID-19 patients reach the point of critical illness where ventilation is necessary, they tend to deteriorate quickly. During the pandemic, patients with other conditions may also present at the hospital needing emergency ventilation. But ventilation of a COVID-19 patient can last for 2–3 weeks. Accordingly, if all ventilators are in use, there will not be time for (...) patients to ‘queue up’ to wait for those who arrived earlier to recover. Those who need a ventilator will die if they do not receive access to one quickly. Many nations now face the prospect that they failed to prepare appropriately for this foreseen risk of pandemic (another issue worthy of legal analysis) and that their populations will need far more ventilators than the health service can supply. For many weeks, the UK has been anticipating a surge of COVID-19 infections that could leave patients queuing in hospitals for ICU care. Luckily, social distancing has ‘flattened the curve’ and this problem might not arise. There is still a degree of risk, however, and other countries will not be so lucky. At some point, during this pandemic or the next, all countries will need to answer hard questions about whether and when scarce ICU resources (such as ventilators, beds and staff) should be either withheld or withdrawn from certain groups of patients solely for the purpose of providing them to others. Attempts to answer these questions can be found in a wide range of ICU triage protocols and ethical guidance documents, many of which embrace the foundational principle of ‘save the most lives’. PurposeTo use the power of knowledge acquisition and machine learning in the development of a collaborative computer classification system based on the features of age-related macular degeneration (AMD).MethodsA vocabulary was acquired from four AMD experts who examined 100 ophthalmoscopic images. The vocabulary was analyzed, hierarchically structured, and incorporated into a collaborative computer classification system called IDOCS. Using this system, three of the experts examined images from a second set of digital images compiled from more than 1000 patients with AMD. Images (...) were annotated, and features were identified and defined. Decision trees, a machine learning method, were trained on the data collected and used to extract patterns. Interrelationships between the data from the different clinicians were investigated.ResultsSix drusen classes in the structured vocabulary were largely sufficient to describe all the identified features. The decision trees classified the data with 76.86% to 88.5% accuracy and distilled patterns in the form of hierarchical trees composed of 5 to 15 nodes. Experts were largely consistent in their characterization of soft, and to a lesser extent, hard drusen, but diverge in definition of other drusen. Size and crystalline morphology were the main determinants of drusen type across all experts.ConclusionsMachine learning is a powerful tool for the characterization of disease phenotypes. The creation of a defined feature set for AMD will facilitate the development of an IDOCS-based classification system. (shrink)

Once upon a time, modal logic was castigated because it ‘had no semantics.’ Kripke, Hintikka, Kanger, and others changed all that. In a similar way, when Relevant Logic was introduced by Anderson and Belnap, it too was castigated for ‘having no semantics.’ The present overview marks a culmination of that effort. The semantic approach described here brings together a number of hitherto disparate efforts to set out formal systems for logics of relevant implication and entailment. It also makes clear (...) (despite some of our hopes and utterances) that the One True Logic does not exist. This is as true for relevant logics as Kripke et al., showed it to be for modal logics. In both cases, subtle (and not so subtle) variations on semantical postulates produce different logics in the same family. The question of which semantical postulates are correct makes no sense without further context, i.e., the questioner needs to answer the question: Correct for what? The question that does remain is: What motivates the relevant family of logics? And this is the question that is the main job for this chapter to investigate. (shrink)

There is no nationalism without some ghost. -Jacques Derrida, "Onto-Theology of National-Humanism" Writing a mere decade ago about Benedict Anderson's Imagined Communities, Timothy Brennan noted that "with the exception of some recent sociological works which use (...) literary theories, it is rare in English to see 'nation-ness' talked about as an imaginative vision--as a topic worthy of full fictional realization.... Even in the underrepresented branch of Third World English studies, one is likely to find discussions of race and colonialism, but not the 'nation' as such" [47]. If these sentences appear astonishing today--if over the past ten years we seem to have done little else but regard the nation as an imaginative construction--our sense of surprise may help to register how rapidly and profoundly Anderson's work has transformed the nature of literary and cultural studies. Viewing the modern nation primarily as an anthropological rather than political category, one that has less inherently to do with ideologies than with kinship, gender, and religion, Anderson argues in Imagined Communities for the irreducibility of material-cultural practices (what he memorably termed "print-capitalism") in creating and sustaining the "imagined community" of a nation whose citizens maintain "deep attachments" to each other in the absence of face-to-face contact: "the members of even the smallest nation will never know most of their fellow-members, meet them, or even hear of them, yet in the minds of each lives the image of their communion" [6]. A Routley-Meyer-style semantics for ConR is given for the system (the completeness of ConR over this semantics is proved in E. Mares and A. Fuhrmann, A Relevant Theory of Conditionals (unpublished MS)). This article responds to Stanley J. Grenz's Templeton Lecture, “Why Do Theologians Need to Be Scientists?” published in the June 2000 issue of Zygon (Grenz 2000). In the first part I outline my reasons for finding the kind of theological reflections in which Grenz engages worthy of attention by noting my disagreement with the view that a sufficient response to theological issues can be formulated on the basis of an examination of our biological nature. I assert, in that connection, the (...) autonomy of reason as a way of investigating and understanding the world. In the second part I respond directly to Grenz by explaining my disagreement with the postmodern critique of science upon which he relies and his adherence to Christian eschatology as an answer to the conundrums into which, he posits, we are drawn as a result of that critique. I note that I agree with Grenz, however, that the activity of valuing is necessarily a forward‐looking Godlike endeavor that is not derivable from science. In the third part I suggest that we must be open to the investigation of the possible existence of an objective realm of value and that, in any case, rejection of the postmodern critique of science in many cases pro‐vides a sound basis for the disciplined resolution of factual questions that frequently lie at the base of disagreements about values. This chapter explores global debunking arguments, debunking arguments that aim to give one a global defeater. I defend Alvin Plantinga’s view that global defeaters are possible and, once gained, are impossible to escape by reasoning. They thereby must be extinguished by other means: epistemically propitious actions, luck, or grace. I then distinguish between three types of global defeater—pure-undercutters, undercutters-because-rebutters, and undercutters-while-rebutters—and systematically consider how one can deflect such defeaters. Lastly, since I draw insights from the literature on perhaps the most (...) widely discussed global debunking argument in the literature, Plantinga’s evolutionary argument against naturalism, I end up responding to many potential problems for it. This includes the so-called conditionalization problem, as well as those raised by Bergmann (2002), Law (2012), Deem (2018), Hendricks and Anderson (2020), and Wielenberg (in Craig and Wielenberg (2021)). This paper begins by tracing interest in emergence in physics to the work of condensed matter physicist Philip Anderson. It provides a selective introduction to contemporary philosophical approaches to emergence. It surveys two exciting areas of current work that give good reason to re-evaluate our views about emergence in physics. One area focuses on physical systems wherein fundamental theories appear to break down. The other area is the quantum-to-classical transition, where some have claimed that a complete explanation of the (...) behaviors and features of the objects of classical physics entirely in quantum terms is now within our grasp. We suggest that the most useful way to approach the emergent/non-emergent distinction is in epistemic terms, and more specifically that the failure of reductive explanation is constitutive of emergence in physics. Sherwood Anderson's Winesburg, Ohio, published in 1919, is an episodic collection of character sketches based mostly around the perspective of George Willard, a small-town journalist who listens to the stories of various characters, often described in grotesque terms, whose passionate inner lives contrast with their limited outwardly lived existences. The initial critical response to these stories was to regard Anderson as a sort of cheap Freudian who was making an obvious criticism of American Puritanism and conformity. One reviewer, (...)
